Detection of heat shock protein 70 (Hsp 70) in choroidal neovascularisation (CNV) secondary to age related macular degeneration (AMD)

Objective
Detection of Hsp 70 in CNV membranes secondary to AMD and its correlation to pretreatment with verteporphin photodynamic therapy (PDT) or transpupillary thermo therapy (TTT).
Methods
CNV membranes were removed by pars plana vitrectomy (ppV) and subretinal extraction. For Hsp 70-detection the streptavidin-biotin method was used. HeLa Cells served as control indicator. Light optical microscopic evalutation was documented with a software tool (DISCUS).
Results
13 membranes were divided into 3 groups: no pre-treatment (11), PDT-pretreatment (4) and TTT pretreatment (2) membranes. The results within these groups were: No pre-treatment group: 6 positive, 2 negative. PDT group: 4 positive membranes. TTT group: 1 positive and 1 negative specimen.
Conclusions
Hsp 70 is present in the human retina in nearly every layer and moreover in the majority of CNV membranes resulting from AMD. A correlation of induction of HSP70 expression to pre-treatment with PDT or TTT on basis of our data could not be verified.
