Holz1 While the knowledge of pathogenetic mechanisms of advanced AMD is rapidly expanding, the initial triggers of the disease are still unknown. A large body of experimental indicates a key role of both oxidative damage and lysosomal dysfunction of the RPE in the early pathogenesis of AMD. Due to its physiological characteristics the outer retina is highly susceptible to lipid peroxidation-mediated damage. We provide experimental evidence that products of lipid peroxidation cause RPE lysosomal dysfunction and thereby induce processes that may be involved in the development of hallmark features of AMD such as lipofuscinogenesis, sub-RPE deposit formation and RPE cell atrophy.
